Research Abstract

BINOLやBINAPに代表される軸不斉ビアリール化合物は、近代の有機化学において欠かす事の出来ない重要な化合物である。これら軸不斉化合物を光学活性体として得るためには、通常、等量以上のキラル源を用いた分割法が利用されてきたが、より効率的に光学活性体を得る手法として、近年、その触媒的不斉合成法の開発が注目を集めている。我々は、軸不斉ビアリール化合物の中でも、その重要性が広く認識されている含窒素化合物に着目し、キラル相間移動触媒を用いた新たなアプローチにより、含窒素軸不斉化合物の触媒的不斉合成法の開発に取り組んだ。まず、代表的な含窒素軸不斉ビアリール化合物であるNOBIN誘導体に着目し、キラル相間移動触媒を用いたN-アリル化反応を利用した速度論的光学分割を検討した。その結果、本触媒反応系を利用する事で効率的な光学分割が可能である事を明らかにした。今回開発した速度論的光学分割は、NOBINのみならず、様々な構造を有するビアリール化合物の不斉合成へと適用可能である事を明らかにした。例えば、ビナフチル骨格だけでなくフェニル-ナフチル、あるいはビフェニルタイプのビアリール化合物の不斉合成にも有効で、いずれの場合も高選択的に生成物を得る事が出来た。また、得られた光学活性ビアリール化合物は、保護基の脱保護を行う事で、容易に目的化合物であるNOBINへと誘導可能である事を示した。また、ここで開発した速度論的光学分割法の知見を活かし、ビアリール化合物の非対称化を利用した、光学活性含窒素軸不斉ビアリール化合物の触媒的不斉合成にも挑戦した。その結果、本法は非対称化反応にも非常に有効であり、目的のビアリール化合物を高エナンチオ選択的に獲得する事が出来た。
