Research Abstract

機能性核酸を疾患治療に応用するためには、核酸を標的疾患組織の細胞の中にまで届ける必要がある。本研究では、肝炎治療や癌治療への応用を目的として肝臓や癌組織へ効率的に核酸を送達可能なドラッグデリバリーシステムの開発を行った。
平成25年度は、in vivo肝臓へ効率よくsiRNAを送達し標的遺伝子のノックダウン(KD)を引き起こすための物理化学的特性のin vivo最適化を行い、50%KD可能なsiRNA投与量ED50を0.06mg/kgまで低下させることに成功した。東京都臨床医学研究所の小原道法教授によって開発されたC型肝炎ウイルス(HCV)に対するsiRNAを搭載したところ、HCV感染ヒト肝臓キメラマウスにおいてHCVの排除と治療効果を示すことが示された(Watanabe T, Hatakeyama H, et al. Scientific Reports, 2014)。本領域公募班の南川典昭教授らによて開発された肝臓特異的microRNA122(miR-122)を抑制可能な核酸を肝臓へ送達し、標的miR-122の抑制とコレステロール低下を誘起することに成功した(Hatakeyama et al. J Control Release, 2014; Takahashi M, et al. Nucleic Acids Res, 2014)。
血中滞留性を付与することで癌組織にsiRNAを送達させ標的遺伝子KDさせることに成功した(Sakurai Y, et al. Mol Ther, 2013). また癌血管内皮特異的なリガンド分子を搭載することで血管へのsiRNA送達と抗腫瘍効果の誘起にも成功した(Sakurai Y, et al. 2014, J Control Release)。本研究により開発されたpH応答性リポソームは肝炎や癌治療への応用が期待される。
