Research Abstract

タウ遺伝子欠失マウスの行動観察からタウが老齢期の認知機能に必要な因子であることを見いだした。さらにMn-MRI観察から若齢期では場所記憶に関して前頭前野、海馬の神経活動が関与し、老齢期になると海馬の神経活動に依存するようになることを見いだした。タウ遺伝子欠失マウスでは老齢期においても前頭前野、海馬の神経活動が観察され、野性型と較べ海馬神経活動の低下が老齢期の認知機能低下に関与すると考えられた。
海馬シナプス可塑性の電気生理学的検討の結果、タウ遺伝子欠失マウスではLTPは野生型マウスと変わりがなかったが、LTD誘導が引き起こされなかった。野生型マウスから得た海馬スライスからタウShRNAによってタウをノックダウンしてもLTD消失が観察され、タウはLTD誘導に必須の蛋白の一つであることが示された。生化学的分画法、電子顕微鏡観察でタウがシナプス領域に存在することが観察され、LTD誘導ではタウの過剰リン酸化を示した。この過剰リン酸化はNMDA受容体インヒビター、GSK-3インヒビターによって抑制されることから、NMDA依存性LTD誘導においてシナプス領域に存在するGSK-3が活性化されタウを過剰にリン酸化すると考えられた。これらの結果から、老齢期における過剰なNMDA依存性LTD誘導が神経変性の要因となることが示唆された(Kimura T, et al.
