Research Abstract

Wnt シグナルは初期発生の体軸や体節、器官形成、細胞極性制御において重要である。近年では様々な体性幹細胞の維持、また分化運命決定に関与する事が注目されている。LGRファミリーのLGR4, 5はWntシグナルを亢進する液性蛋白質のR-spondinsをリガンドとし、Wntシグナルを制御する新たな受容体である。我々はこれまでにLGRsファミリーの一つ、LGR4が乳腺組織形成に果たす機能解析を行った。
Lgr4の遺伝子領域に蛍光蛋白質EGFPを導入したレポーターマウスを用い、乳腺組織に於けるLGR4の発現細胞を同定した結果、乳腺幹細胞を含む乳腺筋上皮細胞でLGR4が発現していた。Keratin5-CreTgマウスを用いたLg4 conditional KOマウス(Lgr4 cKOマウス)の乳腺組織の発生・分化におけるLGR4の機能を解析したところ、Lgr4cKOマウスは乳腺組織のブランチング形成低下を示し、妊娠時において乳腺胞の発達不全から授乳能が失われていた。乳腺上皮細胞は表面抗原CD24, CD49fの発現強度により内腔上皮前駆細胞、基底上皮前駆細胞を分画できる。FACSを用いて10週齢から10ヶ月齢まで様々な週齢のLgr4 cKOマウス乳腺上皮細胞を解析したところ、加齢に伴い細胞分画のパターンが変化して行く事を見出した。内腔上皮細胞画分ではCD49f強発現細胞、すなわち内腔上皮前駆細胞が減少して行く事も見出した。更にマトリゲル上における乳腺上皮細胞の初代培養細胞の三次元培養を行ったところ、LGR4のリガンドR-spondinが乳上皮細胞のコロニーの形成率と枝分かれ構造形成を促進する事を見出した。これらはLGR4が乳腺細胞の未分化性を維持し、乳腺管構造の形成・維持に寄与する事を示す。
