Research Abstract

動植物を問わず多くの生物の受精時において、精子(雄性配偶子)は卵(雌性配偶子)から分泌される物質に対して走化性を示す。この卵に対する精子走化性現象は、体外受精をする多くの動物門において種特異的な反応であることが知られており、精子が最初に同種の卵を認識する分子認証システムである。さらに、この同種の卵を見いだした精子は走化性運動により卵へと誘導され、受精の確率を高めるシステムとして非常に興味深いシステムである。しかし、精子誘引物質に関しては一部の種で同定されているものの、精子-卵由来物質間の分子認証システムについてはまだほとんど何も分かっていないのが現状である。本研究では、精子誘引物質が同定され、その受容体の解明も進んでいるカタユウレイボヤでの成果を土台として、精子がどのように同種の卵由来の物質を認証し、卵の位置を特定し、精子運動制御をしているかを解明することを目的とする。
今年度は、カタユウレイボヤと近縁なホヤ4種について、精子誘引に関する種特異性について詳細に検討し、一部には片方向性の種を超えた精子誘引活性があることが明らかとなった。また、マウスの受精において精嚢分泌タンパク質SVS2がメス体内における生存および機能の維持において必須であることを示し、SVS2が欠損するとin vivoでは子供ができないことが明らかとなった。
さらに、それ以外の種における精子と卵の分子認証システムの基礎研究では、チョウザメおよびホタテ貝について基礎的な精子運動調節機構に関する知見を得ることに成功した。
