Research Abstract

申請者は、ペア抵抗性遺伝子Pit-1/2による植物免疫の誘導機構の解明を試みた。抵抗性遺伝子Pit-1の隣に座乗する遺伝子Pit-2は、Pit-1とタンパク質レベルで直接結合し、免疫受容体複合体を形成することを見出した。ペア抵抗性タンパク質Pit-1/2は異なる機能を持つことを見出しており、Pit-1は恒常的に免疫を誘導するが、 Pit-2 はPit-1の活性を抑制する。Pit-1と Pit-2機能の違いは、スイッチドメインであると考えられているNB-ARCドメインの30アミノ酸の違いにより生み出されることを示唆するデータを得ている。Pit-1を用いた免疫沈降実験から、Pit-1がPit-2以外の複数の抵抗性タンパク質様タンパク質と複合体を形成することも発見した。この結果は、1つの抵抗性遺伝子は、複数の抵抗性遺伝子とタンパク質レベルでペアを形成していることを示唆している。これまで、1つの抵抗性遺伝子が複数の抵抗性遺伝子とペア形成(ペア抵抗性遺伝子群と命名)をする報告はなく、この知見は植物免疫を理解する上で重要な基礎データとなる可能性が高いと考えた。
