Research Abstract

気孔開閉調節に関わる情報変換タンパク質PCaP1および水・CO2透過性アクアポリンPIPに焦点を当て、シロイヌナズナにおけるそれぞれの生理機能を明らかにし、高CO2下での両分子のCO2供給システムにおける役割とそれに関わる量的・機能的調節機構の解明を目的とした。
細胞膜のタンパク質PCaPのN端領域はCa-カルモジュリン(CaM)複合体、ホスファチジルイノシトールリン酸(PtdInsPs)と結合する。PtdInsPsとCaM/Caは拮抗的であり、CaM/Caの結合はPtdInsPsとの結合を解除する。この特性はCaシグナルからPtdInsPsシグナルへの変換を示唆した。2種PCaPのうちPCaP2は根毛形成に関わる。PCaP1は全ての組織で検出されるが、孔辺細胞においては外輪部のみに偏在する。この意味を検討する目的でPCaP1遺伝子のT-DNA挿入破壊株pcap1の表現型を解析したところ、暗所において閉じるべき気孔が閉じないという異常が見られた。また、生理的高CO2条件(700 ppm)で生育させるとpcap1株は野生株に比べて有為な生育向上が見られた。PCaP1は各細胞において独自の多様な役割を果たしている可能性が高く、孔辺細胞での気孔開閉調節は機能の一端であると考えた。分子の特性から解明するためPCaP1のタンパク質構造解析も進めた。
細胞膜アクアポリンPIP1は複数の植物でCO2透過性が認められており、PIP1の量の減少がCO2固定能の低下をもたらす。PIP1とPIP2のT-DNA挿入遺伝子破壊株のうち、pip1;1pip1;2、pip1;2pip2;5の二重遺伝子破壊株はそれぞれ野生株に比べ生育がよい傾向がみられた。PIP1分子種のみならず孔辺細胞で発現レベルの高いPIP2;5もCO2輸送に関わる可能性があり、岡山大学森博士と共同研究を進めてきた。さらに、高温応答性のPIP2;3については、遺伝子上流の高温応答性シス領域の特定、pip2;3株の水分特性、高CO2応答特性を明らかにした。
