Research Abstract

鹿児島県甑島の貝池において紅色硫黄細菌クロマチウムが密集する層の顕微鏡観察，サンプリングおよびそのサンプルを用いた培養実験を行なった。顕微鏡観察において無数の従属栄養性原生生物が確認された。そのうち複数種の繊毛虫類において紅色硫黄細菌様構造が確認された。ただし，その該当構造が共生体由来のものなのか，餌由来のものなのかは判断出来なかった。そのサンプルを紅色硫黄細菌用の培地に接種し培養を行なった。その結果，紅色硫黄細菌と共に特定の原生生物が増殖することを確認した。その原生生物のSSU rRNA遺伝子を決定し系統解析を行なった結果，嫌気性繊毛虫Trimyemaの1種であることが明らかとなった。このTrimyemaの細胞内に特定の共生体由来構造が存在するかどうかを確認するために，その電子顕微鏡観察およびバクテリア/アーキアユニバーサルPCRプライマーを用いた分子検出が現在進行中である。また紅色硫黄細菌様構造は確認されなかったが，今回のサンプリングにおいてストラメノパイル生物であるCiliophrys infusionumの培養株を得た。このC. infusionumのゲノム中には起源の異なる2種類のSSU rRNA遺伝子が存在することを明らかにした。1つのSSU rRNA遺伝子はその祖先から受け継いできた（垂直伝播した）もので，もう1つはパーキンサスと呼ばれる寄生性原生生物から水平伝播によって得られたものであることを系統解析によって示した。これは真核生物で初めて見いだしたrRNA遺伝子の水平伝播の例となる。
