Research Abstract |
本研究では、意思決定に関わる神経回路の網羅的解析とそのための技術基盤の確立を目的とした。具体的には大脳皮質ー視床ー大脳基底核ループにおける直接路と間接路を構成する神経細胞への大脳皮質からの投射様式を標的とし、そのための技術的基盤として光学顕微鏡技術と電子顕微鏡技術の融合による高解像度かつ広範囲に亘るイメージング技術の確立を目指して研究を遂行した。 本年度は昨年度に導入された電子顕微鏡用超薄連続切片自動回収機ATUM(Automated Ultrathin sectioning microtome)を用いて連続切片の自動回収システムを構築した。さらに回収された連続超薄切片の光学顕微鏡での自動画像取得システムの構築を行い、広範囲からの画像取得に成功した。さらに得られた画像を重ね合わせて広範囲の高解像度イメージ像を生成する手法の開発も行った。これら一連の技術的基盤の確立により、広範囲に亘る大脳皮質ー視床ー大脳基底核ループの全体像を高解像度で撮影することが可能となった。直接路および間接路への大脳皮質からの投射解析についての全体像の解明には多くの時間を要するため本研究期間中に完了することはできなかったが、本研究によりその技術的基盤が確立したことにより、その全貌の解明に近づいたと言える。 今後は、本研究で確立した技術をさらに展開させると共に、電子顕微鏡も併用し、シナプスから回路までを範疇としたメゾスコーピックな画像取得およびその解析を目指す。
