Outline of Annual Research Achievements

リビングアニオン重合に基づき、ポリメタクリル酸メチル(PMMA)の両末端にかご型シルセスキオキサン(POSS)ブロックを導入した高分子PPMPを合成した[Mn(GPC) = 28.7k, Mw/Mn = 1.31]。比較のため、PMMA[Mn = 25k, Mw/Mn = 1.09]も試料として用いた。
細胞の接着・増殖特性は、マウス結合組織由来の線維芽細胞L929を用いて評価した。細胞は、無血清もしくは10%ウシ血清(FBS)含有RPMI 1640培地において標準的な培養条件下で培養した。位相差顕微鏡を用いて観察したPMMA膜およびPPMP膜上の細胞接着数および形態を評価した。PPMP膜上に接着した細胞の数は、PMMA膜上のそれと比較して少なく、また、接着面積ならびに伸展率も低かった。これは、疎水性の高いPOSS部が濃縮した膜表面と細胞が強く相互作用するため、分裂および増殖プロセスへの移行が抑制されたことを示している。血清存在下においても、PPMP膜上における細胞は、PMMA膜上におけるそれと比較し、細胞活性が抑制されることがわかった。血小板粘着特性は、正常ヒト全血より調製した多血小板血漿を用いて評価した。各膜は、前処理としてPBS中に所定時間浸漬した後、試験に供した。PPMP膜上に粘着した血小板の数は、PMMA膜上のそれより多く、また、より多くの偽足形成と扁平化が認められた。PMMA膜における血小板数は前処理時間に依らずほぼ一定であるのに対し、PPMP膜におけるそれは、前処理時間の経過に伴い減少した。
これまで得られた成果を総括することで、膜表面における分子鎖凝集状態とその環境応答性が、血小板粘着特性に強く影響を与えることが明らかとなり、POSSを用いた高分子界面設計の設計指針が得られた。
