Outline of Annual Research Achievements

各種有機高分子を共存させた条件でマグネタイト結晶の水溶液プロセス合成を試みた。まず星型高分子を上記の系に添加したところ,マグネタイト結晶形成自体に変化はなかったものの,結晶粒径がやや小さくなる結果となった。これはマグネタイト結晶核への星型高分子の結合により,結晶核成長が抑制されたためと推測される。さらに,らせん状高分子であるDNAを種々の濃度で添加すると,濃度増加につれてマグネタイトの結晶性や生成量が低下し,代わってヘマタイトの生成が認められる傾向にあった。これらの高分子添加は酸化鉄の形態や結晶相に大きな影響を与えることを示している。
さらに,これまでに得られたマグネタイトナノ粒子をもとに微小球を作製するとともに,その薬剤徐放特性を調べた。水溶液プロセスで得られたマグネタイトナノ粒子をW/Oエマルションの水相中に分散させ,これをブタノール中にて脱水凝固させることで,マグネタイト単独,あるいはマグネタイト-多糖類ハイブリッドからなる,直径数~数十ミクロンの真球状粒子が得られた。球状粒子の粒径はエマルション作製時の攪拌速度や界面活性剤濃度によって大きく変化することが分かった。すなわち,速い攪拌速度あるいは高い界面活性剤濃度は,球状粒子の平均粒径を小さくする傾向にあった。骨腫瘍や骨粗鬆症の主要薬剤であるビスホスホネートをマグネタイト微小球に含浸により担持させ,生理食塩水中での放出挙動を調べた。その結果,微小球は7日以上にわたってビスホスホネート薬剤を徐放し続けることが明らかとなった。これは正に帯電しているマグネタイト表面に,負に帯電したリン酸基を有するビスホスホネートがイオン間あるいはイオン-双極子間相互作用により強固に結合したためであると推測される。
