Outline of Annual Research Achievements

外科治療後の組織にプラズマを照射することにより、手術により生じた創傷のすみやかな治癒が観測されている。細胞表面には糖タンパク質等数多くの糖鎖が存在し、細胞表面の保護等の機能を担っている。このため生体へ照射されたプラズマは、まず細胞表面の糖鎖と相互作用してその化学変化を促すものと考えられる。しかし、生体内の糖鎖は高度に不均一であるため、プラズマによる変化を解析することは容易ではない。本研究では、いくつかの構造の明確な糖鎖誘導体にプラズマを照射することにより、その変化を機器分析手段により解析し、プラズマの糖鎖に対する影響を解析する目的で行った。しかし、グルコース、メチルガラクトシド等の糖鎖モデルにプラズマを照射しても大きな構造変化は見られなかった。この結果から糖鎖に多く存在する水酸基に対しては、プラズマは構造変化を引き起こさないことが明らかとなった。
この結果を踏まえ、糖鎖と同様に生体の主要構成成分であるタンパク質に対するプラズマの影響を解析することとした。血中に多く存在するタンパク質としてウシ血清アルブミン、また、他の可溶性タンパク質の例として卵白リゾチームを用いてプラズマ照射を行った。その結果、照射に伴いゲル状の物質の生成が確認された。しかし機器分析により解析したところ、これらのゲル状タンパク質においてもその構造変化は見られないことが明らかとなった。これらのゲルの生成は、プラズマ照射により水の蒸発が局所的に大きくなり、タンパク質の溶解度限界を超えたためと考えられる。
以上の結果から、プラズマ照射により生体物質の化学構造が大きく変化することはなく、主にゲル等の非定常的な物質に対する生体内の血液凝固系が活性化して創傷治癒へと導いているというメカニズムを推定するに至った。
