自閉症関連分子TAO2キナーゼによるスパイン形成制御

Outline of Annual Research Achievements

1)TAO2b遺伝子欠損マウス(TAO2bノックアウト(KO)マウス)の作製:TAO2b調節ドメインをコードするエクソン17-19をloxP配列で挟み、camkII-Creマウスと交配することにより、TAO2bのみ(TAO2aには影響しない)を欠損したマウスを作製した。
2)TAO2bノックアウトニューロン(KO)の形態学的解析:TAO2b KOマウスの脳を薄切後、HE染色し、脳の各部位の形態学的異常を調べたが、明らかな異常は認められなかった。また、DiIをコートした金粒子を遺伝子銃で脳切片へ撃ち込むことにより、海馬ニューロンを標識した。標識された錐体細胞の樹状突起を撮影し、Metamorphを用いてスパインの形態計測を行った。その結果、TAO2b KOニューロンの樹状突起スパインはcamkII-Creのスパインに比べて有意に細く長いことがわかった。
3)電気生理学的解析:TAO2b KOの仔マウス海馬を分散培養し、アストロサイト上でオータプスを作らせた。シナプス形成後、パッチクランプによりEPSC、シナプス小胞数、遊離確率、mEPSCなどを計測し、camkII-Creニューロンと比較したところ、興奮性のシナプス伝達が低下していることがわかった。
4)行動学的解析:TAO2b KOマウスを用いて、水迷路試験やcontextual fear discrimination、social interaction test、three chamber testおよびultrasonic vocalization testなどを行った。すべての試験においてKOマウスの異常が観察された。
