Outline of Annual Research Achievements

脾臓初期発生に必須の転写因子であるTlx1の遺伝子座にCreER-Venusをノックインしたマウスを用いて、Tlx1の発現を指標として脾臓間葉系細胞の起源ならびに機能について解析を行った。その結果、Tlx1発現細胞は脾臓にのみ存在する間葉系細胞であり、線維性細網細胞、濾胞樹状細胞、赤脾髄線維芽細胞などの成熟ストローマ細胞マーカーを発現せず、LTβR、PDGFRα/βならびにCD105陽性の間葉系幹細胞様の性状を示した。また、胎仔期、新生仔期ならびに成体脾臓のTlx1発現細胞の分化運命を追跡した結果、Tlx1発現細胞は、線維性細網細胞、濾胞樹状細胞、辺縁帯細網細胞ならびに赤脾髄線維芽細胞に分化する能力を有することが明らかになった。さらに、ジフテリア毒素誘導的に成体脾臓からTlx1発現細胞を除去した結果、赤脾髄領域の減少ならびに赤芽球・マクロファージの減少が認められ、赤脾髄において赤芽球・マクロファージニッチとして機能することが示唆された。
