Outline of Annual Research Achievements

血管新生は、固形腫瘍増殖や組織損傷時の修復過程に必須であり、その制御は血管内皮細胞の増殖促進と抑制のバランスで成立している。私どもはこれまでに、血管内皮細胞の増殖促進(VEGFシグナル)と抑制(Notchシグナル)のシグナルバランスを制御する分子複合体BAZF-CUL3を同定してきた。さらに、BAZFおよびCUL3のマウス網膜でのノックダウンによる血管新生評価から、BAZFノックダウンに比べ、CUL3ノックダウンでは極めて強い血管崩壊の表現系が現れることを観察した。このことから、CUL3はBAZF以外にも異なる複合体のパートナーが存在し、血管新生に大きく絡む因子であることが強く示唆された。そこで本研究では、CUL3の機能解析をさらに押し進め、血管新生制御の新たな分子機構の解明を目指した。
まず、CUL3の基質選択性はアダプター蛋白質(BTBドメイン・タンパク質:BTBP)によって担われる。ヒトゲノム情報から、BTBPは183種存在することが予測されているが、CUL3のアダプター活性が確認されているのは10種程度にすぎない。そこで、CUL3の基質探索を目的として、ビオチン化CUL3とFlagタグBTBP183種のリコンビナント蛋白質を合成し、アルファスクリーン法によりインビトロ蛋白質相互作用解析を試みた。これまでに、ビオチン化CUL3とFlagタグBTBP174種のリコンビナント蛋白質合成を終了し、アルファスクリーンを施行した。その結果、CUL3との結合活性が確認できたBTBPは100種以上に及ぶことが明らかとなった。また、BTBPの中でもKLHLファミリーに属する51種のBTBPは、全てがCUL3と結合することを確認した。このうち、血管内皮細胞において発現が確認できたCUL3結合活性を有するBTBPは約60種存在することが明らかとなった。今後、これらCUL3-BTBP約60種の基質を探索する。
