新規クラミドモナス突然変異株を用いた中心子構築機構の解明

Outline of Annual Research Achievements

細胞内微小管構造の形成に重要な中心子は複雑で特徴的な構造をもつが、その構築と維持の分子機構はほとんどが未解明である。本研究では、中心子形成に異常をもつ新規クラミドモナス突然変異株の単離・解析によってこの機構の解明を目指した。平成25年度に単離した新規変異株bld13に続き、26年度ではbld13 の新規alleleを単離し、これら2つのbld13変異株(bld13-1, bld13-2)の解析を行った。その結果、以下のことが明らかになった。
1.これらの変異株は、中心子微小管が一部欠失する、中心子から伸長する微小管束の数と配向が異常になるといった、微小管構造と関係する表現型をもつことがわかった。
2.遺伝解析と全ゲノムシークエンスの結果、bld13はγ-tubulin遺伝子に生じた1アミノ酸置換の変異であることが判明した。しかし、意外なことにいずれの変異も大きなインパクトをもつアミノ酸置換ではなく、bld13-1ではThr→Ile、bld13-2ではGlu→Aspであった。
3.変異型γ-tubulin遺伝子を野生株に発現させたところ、変異型の表現型を示すことから、bld13は優性変異であることが分かった。このことは、γ-tubulinが他のタンパク質とともにリング状に会合して微小管重合活性を獲得することと関係があると推察された。
以上の解析により、中心子構造の構築にγ-tubulinに生じた変異が、中心子構造の構築に大きな影響を及ぼすことが明らかになった。
