Outline of Annual Research Achievements

軸糸ダイニンの発現系を繊毛虫テトラヒメナにおいて確立し、軸糸ダイニン複合体内での軽鎖LC1の局在と機能についての研究を行った。軸糸に存在する外腕ダイニンは、3種類の重鎖、2種類の中間鎖、多数の軽鎖から構成される分子量200万に及ぶ巨大タンパク質複合体である。各重鎖はそれぞれ頭部ドメインと尾部ドメインをもち、3つの頭部が尾部で束ねられた三頭構造をとる。中間鎖と軽鎖の多くは尾部に局在することが知られており、軽鎖LC1は頭部ドメインに局在することが示唆されていたが、頭部のどこに局在するのか明らかでなかった。本研究において、外腕ダイニン複合体内での軽鎖LC1の位置を決めるために、軽鎖LC1にHisタグを融合させたタンパク質を、相同組み換えによりテトラヒメナに発現させ、繊毛に局在することを確認した後、組み換体タンパク質を精製し、さらにHisタグをそのリガンドであるNi-NTAを結合させた金ナノ粒子で標識し、電子顕微鏡観察を行った。その結果、LC1は外腕ダイニン複合体中の特定の外腕頭部のストークの先端の微小管結合領域に結合することを明らかにした。さらに、その結合性をストークのキメラタンパク質を用いて調べ、LC1は微小管結合領域のみで十分に結合すること、LC1が結合すると微小管に対する親和性が低下すること、などを明らかにした。軽鎖LC1は真核細胞の繊毛・鞭毛に普遍的に存在し、この変異体はヒトの繊毛病や繊毛形成異常を引き起こすことなどが知られていたが、原因は明らかでなかった。C1が外腕ダイニンの微小管との相互作用を直接コントロールすることを強く示唆するものであり、繊毛運動の機構解明に対して新たな知見を加えた。
