Outline of Annual Research Achievements

マラリア肝細胞期は、ヒトで1週間、マウスではわずか2日間程度で終了し赤内型に移行する。この間無症状であるが、この期間の感染防御にはマラリア原虫特異的CD8+ T細胞による免疫応答が有効であり、マラリアワクチンの有力な標的となっている。我々は、モデル抗原OVAを発現する組換えマラリア原虫とOVA特異的CD8+T細胞を用いて、肝細胞期防御の生体イメージングを行っている。

これまでの研究で、感染肝細胞周囲に抗原特異的CD8+T細胞がクラスターを形成し、原虫排除に導くことを解明した。今回、このCD8+T細胞の抗原特異性について検討した。特異的T細胞としてOVA特異的T細胞受容体(TCR)トランスジェニックT細胞OT-I、非特異的T細胞としてアロ抗原特異的TCRトランスジェニックT細胞2Cを用いた。各々試験管内で活性化後、マウスに受けに移入し、GFPとOVAの融合蛋白を発現する組換えマラリア原虫スポロゾイトを感染させた。約40時間後肝臓の生体イメージングを行ったところ、OT-Iと2C細胞が共にクラスター形成に参画していた。その数は若干OT-Iが多く、OT-I細胞が抗原シグナルを受けている可能性が示唆された。また、2C細胞のクラスター参画は単独移入マウスでは観察されず、OT-I細胞が抗原特異的に活性化されて標的細胞に集まり、その後抗原非特異的に2C細胞がクラスターに参画することが示唆された。

OT-Iや2C細胞のクラスター形成は、百日咳毒素で阻止されることから、ケモカインの関与が示唆されている。樹状細胞やクッパー細胞の関与を含め、肝細胞期原虫排除におけるT細胞のクラスター形成の分子機構は、今後解決すべき課題である。また、クラスターに参画した原虫抗原非特異的T細胞が、マラリア原虫の排除に何らかの役割を担うのかについても今後明らかにしていきたい。
