多次元蛍光イメージングによる下垂体後葉ホルモン分泌・局所神経回路の可視化と光制御

Outline of Annual Research Achievements

バゾプレッシン-eGFPトランスジェニックラットとオキシトシン-mRFP1トランスジェニックラットを交配することによりバゾプレッシン-eGFP×オキシトシン-mRFP1ダブルトランスジェニックラットが得られた。このダブルトランスジェニックラットの視床下部視索上核から大細胞性神経分泌ニューロンを酵素処理後、機械的に急性単離した。蛍光顕微鏡下でeGFP緑色蛍光およびmRFP1赤色蛍光を指標にバゾプレッシンニューロンおよびオキシトシンニューロンを同定し、ホールセルパッチクランプ法により膜電位および膜電流記録を行い、酸感受性について検討した。その結果、バゾプレッシンニューロンの方がオキシトシンニューロンよりも極めて酸感受性が高いことが明らかとなった。
光興奮性タンパク（チャネルロドプシン２（ChR2））-eGFP遺伝子をc-fos遺伝子に挿入した融合遺伝子を用いて作出したトランスジェニックラットに浸透圧負荷（２日間脱水、高張食塩水腹腔内投与）後、視床下部室傍核および視索上核の一部のニューロンにChR2の発現を示すeGFP緑色蛍光陽性のニューロンが観察されたが、機能的な解析（光感受性の検討）までには至らなかった。
また、バゾプレッシン-ChR2-eGFPトランスジェニックラットでは、２％食塩水飲水負荷後に視床下部室傍核、視索上核および下垂体後葉において著明なeGFP緑色蛍光の増加が観察された。さらに共焦点レーザー顕微鏡で観察したところ、eGFP緑色蛍光は大細胞性神経分泌ニューロンの細胞膜に主に限局して発現していた。このラットの視索上核から単離した大細胞性神経分泌ニューロンおよび視索上核を含む脳スライス標本を用いてeGFP緑色蛍光陽性ニューロンからホールセルパッチクランプ法により膜電位もしくは膜電流を記録しながら青色光照射したところ、再現性よく活動電位および内向き電流が記録できた。
