Outline of Annual Research Achievements

自然免疫は微生物の感染に対する生体の初期防御反応である。微生物の構成成分はToll-like receptors (TLRs),NOD-like receptors(NLRs),RIG-I-like receptors (RLRs)などの受容体によって認識されることで様々な免疫応答を引き起こす。ウイルスや微生物由来
の核酸はTLR7,TLR8,TLR9受容体によって認識され,インターフェロンの産生を促し抗ウイルス反応を引き起こす。故に,抗ウイルス薬やワクチンのターゲットとして注目されており,これらTLR受容体によるリガンド認識の詳細機構の解明が待望されている。本申請
課題では,これらウイルス由来の核酸を認識するTLR受容体によるリガンド認識機構やシグナル伝達機構を主にX線結晶構造解析により明らかにする。これは,ウイルスに対する宿主の応答を制御する薬剤の開発に大きく寄与すると考えられる。
本年度は、さらに、ウイルス由来のRNA認識機構を明らかにするために、TLR8とRNA複合体の結晶構造解析を目指した。その結果、TLR8には2か所のリガンド結合部位が存在することが明らかになった。それら2か所の結合部位でRNA由来のリガンドを認識することで活性化することを示すことができた。
