Outline of Annual Research Achievements

本研究は、宿主遺伝子多様性へのウイルス適応機序の解明を目指し、ウイルスと宿主の相互作用が長期間継続し病態形成にいたるHIV慢性持続感染症を対象として、宿主集団内のウイルス感染・伝播におけるウイルスの変化を知ることを目的とした。HIV慢性持続感染症では、細胞傷害性Tリンパ球(CTL)の強いHIV複製抑制圧のもと、ゲノムにCTL逃避変異を有するウイルスの選択が頻繁に認められ、場合によりこの逃避変異がウイルス増殖能の低下に結びつくことも知られている。CTL標的は、MHC-I遺伝子型の違いにより異なるため、HIVは集団感染伝播において複製能確保とCTLからの逃避のバランスのもと変化すると考えられる。本研究では、ヒトHIV感染を反映するエイズモデルにて、SIV伝播実験を行い、ウイルスの変化を検討した。
SIV感染MHC-IハプロタイプA陽性個体からMHC-IハプロタイプE(またはP)陽性個体、さらにP(またはE)陽性個体へと3代の感染伝播(血漿の経静脈接種)を行ったSIVゲノムの解析から、複製能低下を伴うCTL逃避変異も比較的高頻度に維持され、伝播の繰り返しにより変異が蓄積されることが判明した。次世代遺伝子解析技術を用いたウイルスゲノムの解析から、伝播による感染成立時のウイルスゲノムの変化はわずかであり、各個体の慢性持続感染においてウイルスゲノムに変異が蓄積することが確認された。さらに、1代目・2代目・3代目の伝播SIVを血漿より回収し、いずれも培養細胞レベルでの複製能が野生型SIVと比べて低下していることを明らかにした。これらの結果は、多様なMHC-I遺伝子型を有する宿主間の感染伝播におけるウイルスの変化をエイズモデルで実証するものとして重要な成果である。
