Outline of Annual Research Achievements

運動制御における小脳皮質の機能的役割を明らかにする目的で、2光子イメージングを用い、運動課題実行中の小脳プルキンエ細胞の活動解析を行った。まず、小脳皮質の帯状構造を可視化することのできるマウスを用い、小脳皮質の機能単位である微小帯域の機能イメージングを行った。2光子カルシウムイメージングにより、プルキンエ細胞の登上線維応答観察したところ、登上線維応答の同期性と帯状構造がを1細胞レベルで一致していることが明らかとなった。また、覚醒状態のマウスでこの同期性と帯状構造の一致がどのようになるかを調べたところ、帯状構造をまたいで複数の微小帯域が協調して働いていることが明らかとなった。この結果は、小脳皮質微小帯域と登上線維の関係は投射パターンによって厳密に決まっているものの、脳の覚醒状態や行動状態によって、それらの間のカップリングを柔軟に変化させて、情報処理を行っていることを示唆している。次に、微小帯域の運動制御・運動学習における機能的役割を明らかにするために、頭部固定下のマウスで運動課題を学習途中に、GCaMPによるカルシウムイメージングにより、同一個体で経時的に小脳プルキンエ細胞の活動を観察した。その結果、運動開始のタイミング制御に関わると考えられる活動や、運動の結果としての感覚フィードバックを表現していると思われる活動などが観察され、それらの活動が学習の過程で変化すること、運動や感覚情報を担う機能的微小帯域と帯状構造とが一致していることなどが明らかになりつつある。レバー引きなどのより複雑で精緻な多関節前肢運動についても、同様の実験をおこなっており、これらの実験を通して運動制御・運動学習の基盤となる小脳皮質小規模回路の機能的役割を明らかにしていく。
