Outline of Annual Research Achievements

海馬は記憶の形成や想起に関わる脳部位であり、海馬錐体細胞においてグルタミン酸作動性の興奮性シナプスの入力部位である個々の樹状突起棘（スパイン）が記憶に重要であると考えられている。統合失調症等の一部の精神疾患では進行性の記憶障害が生じるが、そのマイクロ病態として海馬でのスパインの萎縮やスパイン数の減少等が明らかにされてきた。しかしこれらのマイクロ病態がどのように進行しているのかは未だに解明されていない。本研究では、マウス海馬スライスの錐体細胞を標本として用い、統合失調症様精神症状を誘発させる薬物の投与や遺伝子の操作によって個々のスパインの形態・機能やシナプス近傍の神経内外環境がどのように変化するのかを明らかにし、記憶障害の発現・進行に関わるマイクロ病態を解明することを目的としている。26年度は前年度に引き続きシナプスの形態とCa2+活動の経時変化をイメージングで測定することを可能にする蛍光プローブの作製を行った。さらにシナプス病態の経時的な進行を明らかにするため、統合失調症様精神症状の誘発との関連が想定されている異常なDisc1変異体等を神経細胞に発現させ、その効果を蛍光イメージングで検討した。一方研究代表者らが以前に開発した赤色Ca2+プローブであるR-CaMP1.07を基にして、超高感度かつ超高速で微弱な神経活動を検出できる赤色蛍光Ca2+プローブR-CaMP2を開発し、従来以上に高精度に神経活動を解析できることを示した。
