Outline of Annual Research Achievements

活性酸素産生酵素NOX1により制御されるうつ様行動発現メカニズムの解明を試みた。
NOX1由来ROSの標的分子の同定:
NOX1由来ROSの標的分子の解析をレドックスプロテオミクスにより行った。慢性コルチコステロン投与によりROS産生上昇が認められるprefrontal cortex (PFC) の野生型マウス(WT)でNR1、NR2B、N-ethylmaleimide-sensitive factor(NSF)のシステイン残基の酸化修飾が認められたが、NOX1遺伝子欠損マウス(NOX1-KO)において、その修飾が抑制された。
NOX1がうつ様行動発現に寄与する神経回路の解析:
慢性コルチコステロン投与によりNOX1由来ROS産生増加がPFCで認められた。しかし、NOX1発現誘導はPFCではなく、ventral tagmental area(VTA)で認められた。これらの結果より、うつ様行動発現においてNOX1が関与する責任神経回路としてVTA-PFCを想定し、NOX1発現を抑制する人工mRNAを組み込んだAAVをVTAに投与することで、この仮説を検証した。VTAのNOX1発現誘導とうつ様行動の発現は正の相関関係が、PFCでのBDNF発現は負の相関関係を示す傾向があり、現在例数を追加して確認を行っている。
