Outline of Annual Research Achievements

現在、腸管には存在する免疫システムが免疫学における新潮流として注目されている。腸管に存在する多種多様な免疫担当細胞が協調的に機能することで、外界から侵入してくる病原微生物に対する生体防御と食餌性抗原や腸内細菌に対する免疫学的寛容という相反する免疫応答を巧みに制御し生体の恒常性を維持している。一方で腸管免疫を介した恒常性維持の破綻は各種免疫疾患の発症につながる。これまでの研究から、腸管免疫の制御における脂質の関与が示されているが、そのメカニズムについては不明な点も多い。本課題においては、代表者がこれまで進めてきたスフィンゴシン１リン酸（S1P）や食餌性脂肪酸を介した免疫制御に関する研究基盤をもとに、腸管組織における各種脂質の産生と認識、さらには免疫疾患との関連を腸内環境因子の関与も含め明らかにするための研究を遂行した。卵を摂取することでアレルギー性下痢を発症する食物アレルギーモデルを用いた検討から、通常用いられる大豆油の代わりに亜麻仁油を含む餌で飼育したマウスにおいてはアレルギー性下痢の発症が抑制されることが判明した。リピドミクス解析や質量イメージング技術を用いた検討から、亜麻仁油に多く含まれるαリノレン酸とその代謝物であるEPAが亜麻仁油で飼育したマウスの腸管組織において増加していることが判明した。さらに脂質代謝物の網羅的解析からEPAの代謝物の一つである17,18-EpETEが亜麻仁油で飼育したマウスの腸管で増加していること、合成した17,18-EpETEを化合物のレベルで投与しても同様の腸管アレルギー抑制効果が認められることが判明し、17,18-EpETEが亜麻仁油を起点に体内で産生される抗アレルギー性脂質の一つであることが明らかになった。
