Outline of Annual Research Achievements

生理活性脂質であるスフィンゴシン1リン酸(S1P)は細胞内で産生された後、輸送体によって細胞外へ供給され、標的受容体に認識されることで様々な生理作用を制御している。これまでに哺乳類では5つのS1P受容体が同定されているが、各S1P受容体の生理機能の特異性や機能的相互作用の全貌解明には至っていない。一方でS1P輸送体は申請者らによってSPNS2が同定されたのみで、その他のS1P輸送体については同定されていない。そこで本研究では新規S1P分泌輸送体の同定及びS1Pの生理機能の解明を目指し、人工ヌクレアーゼであるTALENやCRISPR/Cas9 systemを用いて遺伝子改変ゼブラフィッシュを作成し、その表現型の解析を行う。
TALENやCRISPR/Cas9 systemといった人工ヌクレアーゼはゲノムの標的部位にDNA二本鎖切断を引き起こす。切断部位は細胞の持つ修復機構(非相同末端結合)により修復される際に、切断部位に数塩基の挿入・欠損が高頻度で生じるため、フレームシフトにより標的遺伝子の破壊が引き起こされる。これらの人工ヌクレアーゼを用いて全てのS1P受容体及びS1P合成酵素について遺伝子破壊ゼブラフィッシュを作成したところ、S1P合成酵素の一つであるsphk2変異体においてs1pr2受容体やspns2の変異体と同様の二股心臓の表現型が見られた。spns2やs1pr2変異体と異なり、母親と接合子の両方に由来するsphk2が心臓前駆細胞の遊走制御に必要であった。sphk2 mRNAは発生初期から発現しており、母親から供給されていた。また発生初期における胎児中のS1Pは母親のSphk2に由来することを明らかとした。一方で接合子由来のsphk2は卵黄多核層という胚体外組織に発現していた。これらの結果は卵黄には母親から供給されたS1Pが蓄積しており、この卵黄中S1P及び卵黄多核層で産生されたS1PがSpns2によって胚体へと輸送され、S1pr2受容体に認識されることで心臓前駆細胞の遊走を制御していると考えられる。
