Outline of Annual Research Achievements |
Notchシグナルは、多様な組織の器官発生に関与しており、その異常により疾患を引き起こすことが知られている。従って、その活性調節の仕組みを詳細に知ることは、再生医療、疾患病態の改善、治療に貢献する可能性を持つ。本研究では、Notchシグナルを調節する翻訳後修飾であるユビキチン化とリン酸化に焦点をあて、未解明調節分子機構を明らかにすることを目的とする。本年度は、、1) Notchシグナル受容細胞でのMibユビキチンリガーゼによるNotchシグナル活性化機構について、Mib1の新たなユビキチン化ターゲットを発見した。新たなユビキチン化ターゲットにより、より効率的なNotchシグナル伝達が行われることが示唆された。2)Notchシグナル活性を制御するキナーゼの探索を行い、これまでにNotchシグナル制御に関与することが知られていないキナーゼの同定した。3)細胞移動におけるMib1の機能制御について、Notchシグナルとは独立した機構の存在とそのメカニズム解析を行った。
