Outline of Annual Research Achievements

VOZ(VASCULAR PLANT ONE-ZINC FINGER)1/2は,シロイヌナズナ液胞膜に存在するH+ポンプAVP1遺伝子のプロモーター領域に結合する因子として単離された。 VOZ は,ヒメツリガネゴケから被子植物まで高度に保存されているタンパク質で,in vitroにおいてGCGTNx7ACGC配列に結合し,転写活性化能を持つことから,陸上植物の生存に非常に重要な転写因子であることが予想された。しかしながら,現時点でVOZ1/2は明確な核局在が示されておらず,転写因子以外の機能も考えられる状況である。そこで本年度は,VOZの機能を明らかにするためにストレス環境下におけるシロイヌナズナのVOZ-GFPの動態解析を行った。その結果,通常条件下では細胞質に散在していたVOZ-GFPが,乾燥や高温ストレス条件下では細胞質内で顆粒状構造を形成している様子が観察された。この顆粒状構造は,真核生物細胞がストレス応答時に形成するストレス顆粒または,Processing-body(P-body)マーカーであるtagRFP-PABP8, tagRFP-DCP1と共局在したことから,VOZは,ストレス時にストレス顆粒およびP-bodyに局在することが明らかとなった。これらのことから,VOZはストレス時,特定のRNAの代謝に関与している可能性が示唆された。
