Outline of Annual Research Achievements

1.「CPD光回復酵素遺伝子(PHR)のUVB発現誘導」:イネとシロイヌナズナにおける、CPD光回復酵素(PHR)のUVBによる発現誘導メカニズムに着目した解析を進めた。(1)シロイヌナズナにおいては、UVBおよびUVA/blueによって強く発現が誘導されることを見出した。そして、uvr8, amiR-uvr8cry1cry2, cry1cry2, cry1cry2phyAphyB, cop1, hy5, hyh等の変異体を材料に発現誘導機構を解析した結果、AtPHRは主にUVR8とCryに依存したシグナル伝達系を介して誘導されるが、UVBに対しては、UVR8非依存的に誘導されるシグナル系も有していることを見出した。そしてこのUVR8非依存的に誘導されるシグナル系は、UVBによって誘発されたCPD量とリンクして機能していることを見出した(Li et al. 投稿中)。(2)イネにおいてもOsPHRは、UVBによって発現誘導されるものの、イネにはUVR8ホモログが2つ存在した。そこで、各々、または両者のUVR8の発現を抑制組換え体イネを作製し解析した結果、イネでは各々のUVR8が生体内で機能し、OsPHRの発現を制御していることを明らかにした。
2.「CPD光回復酵素のオルガネラ移行メカニズムとUVB適応戦略機構」:OsPHRの葉緑体移行シグナルは、N末端領域に存在する可能性を見出した。また、核、葉緑体、ミトコンドリアの各オルガネラでのみ機能し得る組換え体を作製し、オルガネラでのDNA修復とUVB抵抗性との関連を解析した結果、どの組換え体もPHRを欠失した変異体と比較してUVB抵抗性を示した。すなわち、植物は全てのオルガネラにPHRを移行する機能を獲得することで、UVB障害を防御する機構を獲得したと考えられた。
