Outline of Annual Research Achievements

葉緑体光定位運動は葉緑体光定位運動は、植物の生存と光合成の効率化に必須の生理現象であり、我々は移動に働く新規アクチン繊維(chloroplast actin filaments、以下cp-actinと略す)構造を発見した。cp-actinは葉緑体の外包膜上に存在するCHUP1により重合・維持される。従って、CHUP1が葉緑体の外包膜上をcp-actin重合部位に向かって移動し、定着するCHUP1の動態制御にはCHUP1の膜との結合、膜からの離脱、膜上の移動の3要素で構成される。CHUP1の細胞膜への結合にはN末端側のcoiled-coilドメイン(CHUP1-NC)を介した細胞膜タンパク質との相互作用が必要であり、その細胞膜局在の因子の一つがCHUP1-Interacting Protein1 (CHIP1)である。さらに、KACとの相互作用も必須である。本年度は、CHUP1の細胞膜からの離脱制御機構を解析した。まず、CHUP1-NCが細胞膜に結合するためにはCHUP1-NC のCHIP1とKACとの相互作用が必要であることを明らかにした。また、phot2欠損変異体ではCHUP1-NCとKAC1が解離できないことから、CHUP1の KACとCHIP1からの離脱にはphot2依存のリン酸化が必要であることが分かった。このことから、phot2依存的リン酸化がCHUP1複合体の結合を解除し、その結果CHUP1が細胞膜から離脱し、移動が始まると考えられた。次に、CHIP1のphot2依存的リン酸化部位を同定した。得られたCHIP1リン酸化部位をアラニンもしくはアスパラギン酸に置換したCHIP1遺伝子を作成し、chip1変異体に導入した形質転換体を作成した。現在、T3世代のホモラインが確立し、CHIP1の分布、葉緑体運動、CHUP1とcp-actinの挙動にどのような影響が出るかを詳細に調べている。
