Outline of Annual Research Achievements

TobがmRNAの安定性を制御するメカニズムを確立するために、以下を進めた。1) 前年度に引き続き、Tobとpoly(A)結合タンパク質(PABP)、CNOT7デアデニレースの3者複合体の結晶構造解析を目指し、それらを大腸菌で発現させることを試みている。
2) Tobは、mRNAの3’UTRに存在するpoly(A)付加シグナルを認識するCPEBと会合すること、更にTob/CPEB複合体はCNOT7と会合し、特定のmRNAを不安定化することが報告されている。そこでTobとCPEBの結晶構造解析を計画した。その為にCPEBと結合するTobの領域(198Ser-236Ser)に含まれるペプチド断片を合成した。今後、このペプチド断片のNMR解析を進めるとともに、全長CPEBを発現させ、Tobペプチドとの結合状態を結晶構造解析する。
3) CPEB/Tob/CNOT7複合体の共結晶解析が進まないことも想定し、Tob/CNOT7が別の3’UTR結合タンパクを探索した。Tob/CNOT7が、特定のmRNA分解に寄与していることを調べる中で、CNOT7欠損マウスが高脂肪食下では肥満になりにくいことを見出した。高脂肪食下CNOT7欠損では、一部の白色脂肪組織(WAT)で熱産生が亢進し、脂肪が蓄積しにくくなっていることがその原因であることが分かった。さらに栄養源からのATP産生を熱産生に代えるUCP1(uncoupling protein 1)の発現が上昇していることを明らかにした。その分子機構を探る中で、CNOT7のみならずTobを欠損するマウスのWATにおいてもUCP1 mRNAが安定化し、発現上昇していることが分かった。さらにUCP1 mRNAの3’ UTRに存在するAU-rich 配列に結合するBRF-1がTobと会合しCNOT7を標的mRNAにリクルートすることを見出した。BRF1とTobの結合領域を決めながら結晶構造解析を進めている。
