Outline of Annual Research Achievements

本研究では、マウス皮膚の発生モデルを用いて、神経-動脈-静脈間の相互作用による併走性制御のメカニズムおよび、これらの併走性がもたらす生理的意義の解明を目的として研究を行った。神経-動脈-静脈が併走する分岐パターンを示すことは古くから認知されているが、その制御機構や、生理的な意義は明確になっていない。併走性について理解を深めることが、血管や神経の秩序ある再構築を必要とする再生医療の実用化には不可欠である。申請者は動脈-静脈間にApelin / APJ signalingが作用し、静脈血管がスライドするように動脈近傍に引き寄せられることを見いだした。まず、Apelin / APJ系がどのような分子機構で、静脈血管の移動を引き起こしているかを網羅的な遺伝子発現変化から検討したところ、顆粒因子としてsfrp1が働いてることが明らかになった。また、移動する静脈血管の周囲では血管を覆う細胞外基質の著しい減少が認められため、マトリックスメタロプロテアーゼの関与を検討した。その結果、Apelin / APJシグナルがsfrp1を介してマトリックスメタロプロテアーゼの産生を誘導していることが明らかとなった。また、動脈-静脈の併走性に異常があるマウスを用いて、併走性の生理的意義を検討したところ、体温調整に寄与している事が明らかとなった。動静脈の相互作用としては、eprhinB2-EphB4系による「反発する力」が作用しており、動静脈の融合を防いでいることが知られていた。本研究の結果から、動静脈の間ではApelin / APJ系による「引きつける力」が働いてい事が解明され、相反する2つの力のバランスによって動静脈の並走構造が形成される事が示された。また、体温維持という生理学的な意義も明らかとなり、血管再生における動静脈構造制御の重要性を明らかにすることが出来た。
