Outline of Annual Research Achievements

大脳皮質においてグリア細胞は神経細胞と並ぶ主要な構成要素である。アストロサイトは脳内に最も豊富に存在するグリア細胞として知られる。大脳皮質(灰白質)のアストロサイトは血管へ仮足を伸ばし、血液脳関門を形成する。また一方で密に分枝した突起を伸ばして相互排他的なドメイン構造を作り、ここに抱えられたシナプスを同期的に調節している。このようにアストロサイトは、脳高次機能に中心的な役割を果たす細胞として認識されるようになった。しかしグリア細胞がどのように移動し、配置されるのかは不明な点が多く、その解明が待たれていた。我々は大脳皮質形成過程における皮質神経細胞の移動様式をライブイメージングする過程で、通常の放射方向に移動する神経細胞以外に、これまで記載の無い奇妙な移動様式を取る細胞の存在に気付いた。これらの細胞は皮質形成過程後期(マウスの妊娠15日目以降)に認められ、最大で120μm/時間という非常に速い速度で、移動方向をランダムに転換させながら皮質内へ移動する。我々は、この移動様式を不軌道性移動と名付けた。様々な細胞系譜解析から、我々は、この不軌道性移動細胞は最終的には皮質アストロサイトへ分化することを明らかにした。さらに我々は不軌道性移動が血管と密接に関わることを観察した。これらの知見は、アストロサイト前駆細胞が活発に動き回る細胞であるという新しい視点を提供する。
