Outline of Annual Research Achievements

神経幹細胞は、大脳新皮質を構築する大量の神経細胞・グリア細胞の全ての起源であり、発生過程に沿って神経幹細胞自身が指数関数的に数を増やす増殖期、神経細胞を産生する時期、胎生後期のオリゴデンドロサイト系譜細胞を産出する時期、次いで胎生後期から生後のアストロサイト産生期と、性質を大きく変化させる。また、胎生期から生後にかけて神経幹細胞の細胞周期は著しく伸長し、成体脳ではほぼ細胞分裂が停止した状態へと移行する。神経幹細胞の分化能力や細胞周期の変化は、大脳新皮質の多様な神経細胞・グリア細胞の産生に深く関わると考えられているが、その分子メカニズムは全くわかっていない。本研究では、神経幹細胞の未分化性の維持に関与し、しかも細胞周期を制御している因子として、Bre1aおよびKlf5遺伝子を取り上げ、その機能解析を行うものである。
平成25年度にはBre1a遺伝子の機能解析を行ったので、平成26年度は主にKlf5遺伝子の機能を解析した。子宮内電気穿孔法を用い、Klf5遺伝子を胎仔期大脳新皮質の神経幹/前駆細胞に過剰発現/機能喪失したところ、神経幹/前駆細胞でKlf5遺伝子の過剰発現によって未分化性が保たれ、細胞増殖が亢進するのに対し、Klf5遺伝子ノックダウンでは分化が促進されることがわかった。これらの結果は、Klf5遺伝子の神経幹/前駆細胞特異的なコンディッショナルノックアウトマウスの解析でも支持された。
