Outline of Annual Research Achievements

昨年度に引き続き、大脳基底核原基からの前脳腹側の様々な領域に分布するアストロサイトが産生されることを見いだした。この領域からは神経細胞も賛成されるため両者の関係に関して更なる解析を行った。具体的には特定の領域の神経細胞とアストロサイトが共通のprogenitor domain(PD)に由来するか否かを検討した。そのため、電気穿孔法を用いて大脳基底核原基にトランスポゼースをコードした遺伝子とGFPをコードする遺伝子を導入した。又一部の実験ではNkx2.1cre:Ai9マウスにこれを導入することでNkx2.1陽性領域(主に内側基底核原基)から産生される細胞を特異的に標識した。その結果、線条体腹側部や梨状葉では、赤色に標識されるNkx2.1陽性領域由来の神経細胞が緑色のみで標識されるNkx2.1陰性領域由来のアストロサイトと密接に接触している様子を確認することが出来た。脊髄では共通のPD由来のニューロンとアストロサイトに機能的連関があるが、前脳では異なるPD由来のニューロンとアストロサイトにも機能的連関があることを示唆している。異なるドメイン由来のアストロサイトにも間興味深い結果が得られ、これらはお互いの領域を避けるかのように重なり合わずに分布していた。
