本研究は、複合適応形質として昆虫の多くの系統で独立に獲得された角(ツノ)に着目する。昆虫を取り巻く様々な環境の中で、繁殖戦略との相互作用を通して進化した新奇適応形態である角には、他の生物群に比類のない極めて多様な形態が存在する。本研究では、申請者がこれまでに確立した遺伝子機能解析法を生かし、主に次世代シーケンサーを用いた比較トランスクリプトーム解析により角形成を制御する遺伝子を同定し、角形成遺伝子ネットワークの進化メカニズムを解明する。 カブトムシの角原基で発現する遺伝子の転写開始点を網羅的に同定するため、新学術領域研究「ゲノム支援」のサポートにより、雌雄それぞれ頭部と前胸部の角原基をサンプルに用いてオリゴキャップ法に基づくTSS-seq解析を行った。その結果、各サンプルのリード数は48 M~123 Mと良好な結果が得られた。 テトラサイクリンOFFシステムを行うpiggyBacベクター(pBac[Dm-hsp70-tTA, 3×P3-ECFP] およびpBac[tetO-EGFP, 3×P3-DsRed])を用いて、藤原班により開発された新規 in vivo エレクトロポレーション法の確立をカブトムシにおいて試みた。その結果、2齢幼虫を用いた場合、高い確率でGFP蛍光の発現が検出され、カブトムシにおいても本法の有効性が確認できた。
