Outline of Annual Research Achievements

抗がん剤への腫瘍応答性を規定する機序として、腫瘍内ミエロイド細胞が、抗がん剤刺激によるストレス下にて「癌幹細胞誘導・活性リガンド」発現を誘発することで、Paracrineに癌幹細胞による発癌活性および治療抵抗性の獲得に貢献するという仮説を検証するために、抗がん剤耐性下での癌幹細胞によるミエロイド細胞機能修飾能に着目した。抗がん剤耐性がん細胞から分離された癌幹細胞では、抗がん剤感受性癌幹細胞と比較してNF-κBやIRFなど恒常的な炎症シグナル活性が誘導されていること、とりわけ発がんマクロファージ誘導に重要なサイトカインであるマクロファージ・コロニー刺激因子(M-CSF)の高い産生能を有することを同定した。このM-CSF産生に焦点を当ててその制御に係わる転写因子をスクリーニングしたところ、IRF5が重要な制御因子であることを同定した。さらに、抗がん剤耐性がん幹細胞は抗がん剤感受性癌幹細胞や非癌幹細胞に比してIRF5を高発現していること、IRF5干渉RNAにより癌幹細胞からのM-CSF産生が著名に抑制されることが判明した。さらに、IRF5は癌幹細胞からのM-CSF産生を介して、マクロファージ腫瘍内浸潤や、CD163+CSF1受容体+M2マクロファージへの分化誘導に重要な役割を果たしていることが明らかになった。さらに、このM2マクロファージは、非がん幹細胞の自己複製能や抗がん剤抵抗性を誘導することで、癌幹細胞様形質誘導に貢献していた。以上より抗がん剤耐性下においては、がん幹細胞はIRF5-M-CSF経路を介してM2マクロファージを誘導することで、発癌活性の増強に重要な役割を果たしていた。以上の解析により、微小環境中の「免疫学的ニッチ」と癌幹細胞活性の分子連関に関して新たな知見を得た。
