マウスやラットでは、出生期に確定した脳の性は一生涯変わることがないのに対し、魚類の脳は生涯にわたって、性を変えるポテンシャルをもっている。そこで本研究では、脊椎動物における脳の性の確かさと揺らぎやすさの分子基盤、そして、そこに存在する基本原理を理解することを目的として、魚類(メダカ)の脳における性差構築と、その可逆性のメカニズムを解析することとした。本年度は、メダカの脳内の性行動中枢(終脳腹側部の神経核であるthe supracommissural/posterior nuclei of the ventral telencephalic area(Vs/Vp)および視索前野の神経核であるthe magnocellular/gigantocellular portions of the magnocellular preoptic nucleus(PMm/PMg))でメス特異的に発現する神経ペプチドneuropeptide B(NPB)の機能解析を進めた。ゲノム編集技術の一種であるtranscription activator-like effector nucleases(TALEN)を用いたNPBとその受容体のノックアウト個体を作出し、表現型解析を行った結果、NPBシグナルは、メスの性行動に際して、オスの受け入れを抑制する機能をもつことが示唆された。このNPBシグナルはメスの性行動中枢だけに存在する機構であるが、この機構によって、メスは、オスと出会ってすぐ産卵するのではなく、求愛ダンスのうまさなどの情報を得た上で、より優れたオスを選択しているのかもしれないと考えられた。
