本年度は、白金系抗がん薬の細胞内蓄積におけるOCT3とMATE1の役割解明と細胞死誘導を制御する情報伝達経路の分子生物学的役割を解明し、それらを統合したモデルの構築を目指し、in vitroおよび動物実験系を用いた検討を行った。その結果、筋肉痛を起こしやすいオキサリプラチンは広範囲に発現するオOCT3によって取り込まれる一方、マウスにおいては排出トランスポータMATE1の寄与が小さいことが明らかとなった。さらに、腎臓においては腎毒性を引き起こすシスプラチンと腎毒性を起こさないオキサリプラチンがほぼ似た挙動を示す一方、腎毒性誘発に関わるPKCdeltaの活性化が大きく異なり、この違いによる尿細管構造維持がシスプラチンの陣毒性発現に寄与することを示した。これらより、取り込みトランスポータOCTと排出トランスポータMATEの寄与による細胞内蓄積と、それに引き続く細胞毒性メカニズムを明らかにし、副作用発現を予測するシミュレーション構築に繋がったと考える。
