Outline of Annual Research Achievements

ミトコンドリアはATP産生以外にCa2+貯蔵・制御器官としても機能する。ミトコンドリアからのCa2+放出は内膜のNa+/Ca2+交換体（NCX）により行われるが、虚血時など細胞内Na+濃度が上昇するような条件下では、ミトコンドリアNCXと細胞膜NCXが協調的に細胞内Ca2+を増加させる方向に働くことが推測される。また、ミトコンドリアからのCa2+放出は筋小胞体のCa2+再充填に寄与すると考えられ、細胞膜、ミトコンドリアおよび筋小胞体間の動的Ca2+制御は心機能維持や心不全発症機序に重要であることと想定されるが、統合的理解には至っていない。本研究では、これらミトコンドリアNCXの分子実体NCLXの発現を増減させた遺伝子改変マウスと、申請者が既に保有しているNCX1遺伝子改変マウス（遺伝子欠損および心筋特異的高発現）との相互交配により、両者の相対的イオン輸送バランスを変えたモデルマウスを作出し、ミトコンドリア－細胞膜間の統合的Ca2+制御の心機能維持における役割を個体レベルで解析し、その制御異常と心不全発症の機序の解明を目指す。
平成26年度は、前年度に得られたNCLXホモノックアウトマウスの心機能について、カテーテルを用いた左心室圧測定などさらに詳細に検討したが、野生型マウスとの差は見られなかった。しかしながら、単離心筋細胞においてカフェインによる細胞内Ca2+上昇がNCLXホモノックアウトマウスで低下しており、ミトコンドリアでのCa2+放出の異常により、筋小胞体のCa2+が減少する可能性が示された。また、NCLXホモノックアウトマウスを用いて圧負荷モデルを作製したところ、大動脈結紮モデルでは心肥大形成および心機能低下が抑制される傾向が見られた。
