細菌の細胞分裂ダイナミクスの構造機能相関解析

Outline of Annual Research Achievements

FtsAの結合によるFtsZへの影響を活性測定(酵素カップリング法と化学的リン酸定量法)によって確認したところ、FtsAの結合によってFtsZの活性が上昇することがわかり、協調現象を初めてアッセイで示すことができた。さらに、蛍光エネルギー移動(FRET)を利用してタンパク質-タンパク質相互作用(PPI)を阻害する化合物の探索を行った。その結果、10種を超える化合物が得られたが、再現性が乏しかったため現在活性測定法などから検証している。さらに、黄色ブドウ球菌(MRSA)のFtsZのX線構造解析を行ったが、FtsZは同一結晶の中に、大きく構造が違う2種(構造A, B)で存在していることがわかった。結晶化条件は生理的条件に近い条件(pH 7.5, 100mM NaCl)であることから、FtsZは溶液中で大きく揺らいでいることを示唆していた。これは、FtsZで初の発見である。別途、個々で得られたFtsZの結晶を、新規阻害剤(米国ラトガース大のD. Pilch教授らが開発してきたもの)を含む溶液にソーキングして解析したところ、その阻害剤は構造選択的に結合していた。この結果は、FtsZの構造選択的阻害剤の可能性を示唆している。
