Outline of Annual Research Achievements

本研究では、細胞内生体分子を時間と空間を制御して可視化し、細胞レベルでの機能解明に取り組んだ。蛋白質の機能解析では目的蛋白質を蛍光蛋白質と融合発現させて蛍光観察する手法が一般的であるが、発現タイミングや発現強度の制御は容易でなく、詳細な時間と局在解明に対応した技術を創り出す研究は皆無であった。そこで、蛍光プローブを目的蛋白質に集積化させる技術の開発を進め、細菌由来の酵素(β-lactamase)変異体(BL-tag)を使った新規蛋白質ラベル化法を開発した。
さらにこのラベル化法を用い、細胞内のpH低下部位のみを可視化できる技術へと応用展開した。具体的にはpHに応答した蛍光OFF/ONスイッチ機能と、標的タンパク質に特異的に集積化する機能を兼ね備えた分子プローブを設計、合成した。このプローブは細胞内の蛋白質に特異的に集積し、細胞内低pH環境下の蛍光イメージングに利用できることが判明したため、このプローブを使ってオートファジーのイメージングを行った。オートファジーは細胞が飢餓状態に陥ると発揮される、分子そして細胞細内小器官の分解過程である。これまでに、蛍光蛋白質を用いてオートファゴソーム(分解系)形成をイメージングする試みは行われてきたが、実際に分解系が機能するタイミングを可視化することはできなかった。本アプローチでは、分解系を形成する蛋白質にpH感受性プローブを集積化することにより、分解系が形成されると初めて発蛍光する実験系を構築することに成功した。機能分子(pHプローブ)を発現場所を限局した蛋白質に組み込むことで初めて可能となる可視化手法であり、経時的にオートファゴソーム形成を初めてイメージング出来ることを示した。
