Outline of Annual Research Achievements |
分子複合体の構造解析は、多くの場合、X 線結晶解析、NMR、電子顕微鏡、により行われている。本研究では未変性状態での質量分析(Native MS)により複雑な分子複合体の形成機構の解明を行い、分子複合体形成機構の解明を行うことを目的とした。研究対象としては、20S プロテアソームのαリングを形成するサブユニットの内、α6、α7 サブユニットに着目して研究を行った。まず、それぞれのαサブユニットについて、Native MSを行った。その結果、α6サブユニットは、ほぼ単量体として存在しており、α7サブユニットは14量体を形成していることが明らかとなった。α7サブユニットの14量体形成については、結晶構造解析との結果と一致した。その後、α7サブユニットに対して、α6サブユニットの添加実験を行った。その結果、α6サブユニットは、α7サブユニットに直接結合することが明らかとなった。さらに、α6サブユニットの結合により、α7サブユニットの14量体は二つの7量体に解離し、単量体のα6サブユニットと結合することが明らかとなった。これらの結果より、以下のモデルの提唱を行った。α7サブユニットは、溶液中で7量体と14量体との平衡関係にあるが、この結合は非常に強く、ほぼ14量体を形成している。そこにα6が添加されると、14量体の形成が不安定化され、7量体に解離し、α6-α7ヘテロ8量体を形成する。この成果により、プロテアソームの形成には、各サブユニットの結合反応が関わっているのではなく、各サブユニット複合体が、ダイナミックな離合集散を通じて関与することが明らかとなった。これらの成果は、Scientic Reportに掲載された。
