Outline of Annual Research Achievements |
細胞膜の受容体蛋白質が細胞外情報に応答して動的に会合体の組み替えを起こし、細胞内外の蛋白質分子との特異的な反応場を形成する過程の理解を目標として研究を行っている。細胞膜における、上皮成長因子受容体(EGFR)・代謝型グルタミン酸受容体(mGulR)の1分子動態を研究対象とする。本年度は主として、mGluRの動態解析に取り組んだ。 mGluRは3量体G蛋白質共役型受容体(GPCR)の一員であり、記憶・学習形成や統合失調症に関与する。グルタミン酸を生理的アゴニストとして活性化されるが、その他種々の人工的アゴニスト、アンタゴニストや調節分子が知られている。mGluRは、細胞外ドメインで共有結合した恒常的2量体を形成している。Halo-tag分子を融合したmGluRを培養細胞に発現させて蛍光標識し、細胞膜における運動・会合の1分子計測を行った。mGluRは複数の運動状態間を遷移しており、2量体がさらに集合した高次会合体を形成することが明らかになった。mGluR会合体の示す拡散状態のうち、最も速い状態はG蛋白質との相互作用、最も遅いものはコーテッドピットとの相互作用に関係する。平均側方拡散係数はアゴニスト、アンタゴニスト、ネガティブアロステリックモジュレータによって変化し、mGluR分子の拡散運動と活性化状態の間に定量的な関係があることが示された。
