Outline of Annual Research Achievements

新学術領域「天然物ケミカルバイオロジー」の名の下、本研究では、大腸菌由来の再構成型無細胞翻訳系（PUREシステム）と、分子進化工学的手法の一つであるmRNAディスプレイ法を組み合わせ、無細胞発現システムHUPEX（Goshima et al., Nat. Methods, 5, 1011, 2008）等により調製されたヒトタンパク質を標的として、天然物を模したペプチドの大規模ライブラリーから分子標的リガンドを探索する方法を開発した。その結果、HUPEXにより調製されたヒトタンパク質を標的とする、新規環状ペプトイド化合物を同定することに成功した。
また、本領域の副題「分子標的と活性制御」の考えに基づき、クリーンルーム内設置型の超高感度質量分析システムDNLC-MS/MS（Natsume et al., Anal. Chem., 74, 4725, 2002）を用いたプロテオミクスアプローチにより、同定した分子標的リガンドのタンパク質・タンパク質間相互作用の阻害部位の同定（分子プロファイリング）を可能にする技術の開発を行なった。その結果、同定した環状ペプトイド化合物がタンパク質・タンパク質間相互作用を阻害していることを実証することに成功した。
更に、新学術領域内の連携研究として、班長の入江一浩教授との共同研究により、アルツハイマー病に関連する（化学合成された）アミロイドβタンパク質を標的とする天然物様ペプチドリガンドの大規模ライブラリーからの探索も推進した。
