Outline of Annual Research Achievements

平成27年度は、既に確立している収束的合成経路に従って、糖を有するカルデノリドとして19-ヒドロキシサルメントゲニンラムノシド、ブリオフィリンCの類縁体となる非天然型ブファジエノリド、C17-エピブリオフィリンCの2つの化合物を合成する事に成功した。具体的には、既に合成を完了している19-ヒドロキシサルメントゲニンから、第一級ヒドロキシ基をブロモベンゾエート、第二級ヒドロキシ基をアセテートとして保護した後、トリクロロアセトイミデートを脱離基として有しヒドロキシ基をベンゾエートとして保護したラムノース誘導体とのグリコシル化により、糖を導入した。その後、全ての保護基を塩基性条件下にて除去し、19-ヒドロキシサルメントゲニンラムノシドを合成した。また、前年度までに確立した方法によりブリオフィリンCの骨格を構築した後、C16-17二重結合を水素添加によって還元した。次いで、ヒドロキシ基の保護基として用いたTBS基を除去し、C17-エピブリオフィリンCを合成した。還元の立体選択性を逆転させてブリオフィリンCを合成する事を目的とし、アンドロステロンから容易に導ける化合物を用いて、モデル研究を行った。既にWisnerらによって報告されている結果を基にして、ピロン等価体であるフランを導入してC16-17二重結合を水素添加したところ、望む立体選択性で反応が進行する事が分かった。これによって、ブリオフィリンCの合成につながる知見を得ることができた。
