Outline of Annual Research Achievements

本研究では、生理活性天然物が結合する標的タンパク質、あるいはその複合体に対して、細胞内で化学修飾する方法の開発を行い、そのまま抽出／同定することを目標とする。前年度までに用いてきた手法は、前所属先で開発されたAGD化学で、この方法は、タンパク質に認識される小分子リガンドに、有機触媒であるDMAPを連結したリガンド連結DMAPと、DMAPに活性化されて反応するチオエステル誘導体の２種類を用い、標的タンパク質の近傍でのみチオエステルをDMAPで活性化することで、標的特異的なラベル化が進行する。本反応を生きた細胞の中で行うための足がかりとして、前年度までに細胞内在性のモデルタンパク質を標的としたラベル化を達成してきた。本年度は、本手法を細胞表層膜タンパク質に適用し、その抗体にAGD化学を適用することで、抗原・抗体反応に基づく特異的なラベル化に成功した。この成果は我々が独自に開発したリガンド指向性化学に用いるリガンド分子を抗体に拡張できた点で、あらゆる標的をもラベル化できる可能性が高まり、一般性の著しい拡張に繋がるものである。一方で本年度は、植物細胞においてもタンパク質ラベル化を適用し、特に植物ホルモン受容体を標的としたラベル化、および天然化合物リガンドの未知標的タンパク質へのラベル化・同定を検討した。その結果、植物細胞の細胞分画等の条件の最適化を経て、標的の検出まで達成することができた。特に植物細胞では、ラベル化試薬等の機能性分子の細胞内導入率の向上、細胞内局在等の物性制御などが課題として浮かび上がってきており、今後さらなる発展が期待できる重要な知見が数多く得られた。
