Outline of Annual Research Achievements

TRPチャネルは細胞内外の環境変化を感知して細胞内シグナルに変換する分子センサーとして働くため、様々な創薬ターゲットとして期待されている。ヒトには27種類ものTRPチャネル種が存在するが、それらが別々に、異なる刺激に対して応答するため、様々な疾患発症治療の鍵因子となっている。電子顕微鏡解析と計算に基づく単粒子解析法を主な解析技術として、TRPチャネルと天然物リガンドの複合体構造解析を推進した。結合動態の詳細な可視化を行い、多彩な天然物リガンドに応答するTRPチャネルのマルチシグナル活性化機構の解明研究を行った。
領域内の上杉らのグループと協力して、ワサビ成分の受容体であるTRPA1受容体について共同研究を遂行し、負染色電子顕微鏡画像から得られたTRPA1の構造に対し、TRPA1に特異的に作用する新規リガンドの結合可視化を行った。電子顕微鏡解析から得られた結合ドメインは、生化学実験の結果ともよく一致しており、両データを合わせることで結果の信憑性を高めることができた。成果をJACS誌に報告した。またNMRを主たる解析手法として用いている高橋らのグループとの領域内共同研究では、界面活性剤に可溶化した１回膜貫通型タンパク質C99と可溶化された脂質２重膜をMSPが取り囲んで安定化させたＮLPの電顕解析を行った。結果を日本薬学会で報告した。これらの領域内連携を通じて、新しい研究分野の開拓に努めた。分子ラベリング技術の開発では、電子顕微鏡解析には金粒子ラベル法が一般に用いられるが、特異抗体Fabや小分子化されたアビジン分子を用いて、画像処理上で結合した小分子を提示する方法の検討を行った。
また本技術の普及を目的に広く研究者が目を通す科学雑誌に概説を記し、さらに新たに本分野を目指す研究者を対象に、英文での入門書の執筆を行った。
