Outline of Annual Research Achievements

近年、水の中での触媒反応が注目を集めている。従来のLewis酸触媒は、微量の水でも触媒能が失活していたが、ランタノイド(Ln)や一部の遷移金属(TM)を含む錯体は、水中でも高い触媒活性を持つことが知られている。また、水中で高立体選択的に生成物を与える不斉触媒の報告も相次いでいる。しかし、水中で高い立体選択性を与える不斉触媒の配位子は、有機溶媒中での反応において、立体選択性をほとんど発現しなかったような分子が多い。水の有無によって、高い立体選択性の発現を可能にする触媒が異なるのは、何故だろうか?
この問いに答えるため、平成26年度は(i)不斉DOTA誘導体配位子を持つLn(III)触媒を用いる水中での向山アルドール反応の機構について調べ、平成27年度はこれに引き続き、(ii)不斉ビピリジン誘導体配位子を持つ鉄(II)触媒を用いる同じ反応の立体選択性発現機構を調べた。
向山アルドール反応の場合、立体選択性を決定するのは、反応の律速段階であるC-C結合生成段階である。そこで、この段階の遷移状態を、「決め打ち」ではなく、自動反応経路探索(GRRM)の一つ、人工力誘起反応(AFIR)法を用いて網羅的に探索したところ、定性的に立体選択比の実験値を再現することができた。
(i)と(ii)の触媒を用いる反応の解析結果を比較すると、以下のことを見出すことができた。触媒の構造は(i)は非常に柔らかく複数のコンフォマーが共存していたが、(ii)は硬い構造を持つため、コンフォマーは一つだけしか存在しなかった。しかし、(i)(ii)は両者とも、C-C結合生成に伴い、基質の触媒に対する配位構造が柔らかく動くことが、立体選択性発現の鍵になるという共通点を持つことが分かった。
