Outline of Annual Research Achievements

実際にタンパク質が機能を発現する細胞内は，生体高分子やイオンなどが極端に混み合った環境にある．本研究では，生体高分子の立体構造安定性に対して，「分子の混み合い効果」，「電解質の添加効果」，「細胞の大きさ（閉じ込め効果）」などの環境因子がどのような影響を与えるかに注目するとともに，細胞の大きさと同程度の微小空間領域における光と分子の相互作用の特異性を利用した高感度顕微分光法の確立を目指した研究を行った．
①リボヌクレアーゼA (RNaseA) に対する電解質および分子混み合いの複合効果：RNaseA水溶液にホフマイスター塩であるNaSCNとポリエチレングリコール (100 mg/mL)を同時に添加すると，それぞれを独立に添加したときの融解温度の変化の和とはならないことが分かった（複合効果）．昨年報告したポリ(N－イソプロピルアクリルアミド) の結果と比較することによって，高分子への電解質の吸着の大きさと複合効果の大きさには相関があることが明らかとなった．
②単一微小液滴内での色素分子間のフェスルター共鳴励起エネルギー移動 (FRET)：イオントラップを用いて，色素分子が溶存した単一微小液滴 (直径約10m) を空間捕捉し，色素分子間のFRETを観測した．バルク溶液における通常の蛍光を観測する方法と比べて，微小液滴からのレーザー発振を観測することでドナー－アクセプター間のFRETを高感度に観測できることを明らかにした．
③単一微小液滴表面に形成された単分子膜のレーザー顕微分光：イオントラップを用いて空間捕捉した単一微小液滴表面に色素標識された界面活性分子からなる単分子膜を形成し，レーザー顕微分光を行った．微小液滴界面に存在する色素分子は，液滴界面に形成される電磁場モードと強く相互作用するため，極めて低いしきい値でレーザー発振に至ることが明らかとなった．
